Adrenomedullin–Receptor Activity-Modifying Protein 2 System Ameliorates Subretinal Fibrosis by Suppressing Epithelial-Mesenchymal Transition in Age-Related Macular Degeneration

چکیده
Age-related macular degeneration (AMD) is a leading cause of visual impairment. Anti–vascular endothelial growth factor drugs used to treat AMD carry the risk inducing subretinal fibrosis. We investigated use adrenomedullin (AM), vasoactive peptide, and its receptor activity-modifying protein 2, RAMP2, which regulate vascular homeostasis suppress The therapeutic potential AM-RAMP2 system was evaluated after laser-induced choroidal neovascularization (LI-CNV), mouse model AMD. Neovascular formation, fibrosis, macrophage invasion were all enhanced in both AM RAMP2 knockout mice compared with those wild-type mice. These pathologic changes suppressed by intravitreal injection AM. Comprehensive gene expression analysis choroid LI-CNV or without administration revealed that fibrosis-related molecules, including Tgfb, Cxcr4, Ccn2, Thbs1, down-regulated In retinal pigment epithelial cells, co-administration transforming factor-? tumor necrosis factor-? induced epithelial-mesenchymal transition, also prevented Finally, C-X-C chemokine type 4 (CXCR4) inhibitors eliminated difference fibrosis between findings suggest suppresses suppressing transition. blindness, global prevalence about 8.7%.1Wong W.L. Molecular mechanisms of subretinal fibrosis in age-related macular degeneration.
Subretinal fibrosis is a result of a wound healing response that follows choroidal neovascularization in neovascular age-related macular degeneration (nAMD). Although anti-vascular endothelial growth factor therapy has become a standard treatment that improves visual acuity in many nAMD patients, unsuccessful treatment outcomes have often been attributed to the progression of subretinal fibrosi...
متن کاملEmodin ameliorates bleomycin-induced pulmonary fibrosis in rats by suppressing epithelial-mesenchymal transition and fibroblast activation
Aberrant activation of TGF-β1 is frequently encountered and promotes epithelial-mesenchymal transition (EMT) and fibroblast activation in pulmonary fibrosis. The present study investigated whether emodin mediates its effect via suppressing TGF-β1-induced EMT and fibroblast activation in bleomycin (BLM)-induced pulmonary fibrosis in rats. Here, we found that emodin induced apoptosis and inhibite...

Introduction: Age-related macular degeneration (AMD) is one of the major causes of visual loss among the elderly. It causes degeneration of cells in the macula. Early diagnosis can be helpful in preventing blindness. Drusen are the initial symptoms of AMD. Since drusen have a wide variety, locating them in screening images is difficult and time-consuming.
